Stanley Wells taught at the Shakespeare Institute of the University of Birmingham from 1962 till 1977, when he became head of the Shakespeare Department of Oxford University Press with the responsibility of producing a new Oxford edition of the Complete Works. This important and innovative edition appeared in 1986. He remains General Editor of the multi-volume Oxford Shakespeare editon. While in Oxford, he held a Senior Research Fellowship at Balliol College. In 1987, he returned to the Shakespeare Institute as Director and Professor of Shakespeare Studies. Since retiring from this position he has devoted himself full time to editorial and authorial work on Shakespeare.

He holds honorary doctorates from Furman University, South Carolina (1975) and the University of Munich (1999). In 1995 he was awarded the Walcott Award of the Library Association for London, where he took his first degree. He is active as Chairman of the Shakespeare Birthplace Trust, Vice-Chairman of the Governors of the Royal Shakespeare Theatre, Chairman of the International Shakespeare Association, and a Trustee of the Globe and Rose Theatres.
